With a relatively small bank of donors, we can provide an "off-the-shelf" CAR or TCR-T cell solution for a large proportion of the population.Ĭonclusions. Importantly, these allogeneic T cells had comparable functional activity to their autologous T cell counterparts in in vitro assays (tumor cell killing and cytokine release) as well as in vivo tumor models. Using our proprietary T cell engineering process, we successfully generated allogeneic T cells with sequential KOs and insertion of a tumor-specific TCR or CAR with high yield. TRAC edited donor T cells did not induce GvHD in an immune compromised mouse model over the 90-day evaluation period. Edited T cells were protected from host NK cells, both in vitro and in an in vivo model engrafted with functional human NK cells. By knocking out select HLA class I and class II proteins, we were able to avoid host CD4- and CD8-T cell-mediated recognition. Alloreactivity by CD4 and CD8 T cells, NK killing, GvHD induction and T cell function was assessed in vitro and/or in vivo. An AAV-mediated insertion of a CAR or TCR into the TRAC locus is used in parallel with the TRAC KO step to redirect the T cells to tumor targets of interest. Additionally, we utilize potent TCR-α and -β constant chain ( TRAC, TRBC) gRNAs that achieve >99% KO of the endogenous TCR, addressing the risk of GvHD. To solve the issue of rejection by alloreactive CD4 and CD8 T cells, we knocked out (KO) select HLA class I and class II expression with a sequential editing process. Here, we developed an immunologically stealth "off-the-shelf" T cell strategy by leveraging our CRISPR/Cas9 platform and proprietary sequential editing process. Because activated T cells upregulate HLA class II, rejection by alloreactive CD4 T cells should also be addressed. To overcome this, researchers are exploring insertion of the non-polymorphic HLA-E gene, which can provide partial but not full protection from NK cell-mediated lysis. However, loss of HLA class I sends a "missing-self" signal to natural killer (NK) cells, which readily eliminate B2M null T cells. Other strategies include deletion of the B2M gene to remove HLA class I molecules and avoid recognition by host CD8 T cells. Although showing responses in the clinic, this approach carries the risk of infections and the durability of the adoptive T cells is uncertain. Some approaches utilize prolonged immune suppression to avoid immune rejection and increase persistence. The recognition of allogeneic cells by the host is a complex issue that has not been fully solved to date. Alloreactive T cells from unrelated donors can cause graft versus host disease (GvHD) for which researchers have successfully used nucleases to reduce expression of the endogenous T cell receptor (TCR) in the allogeneic product. There is a strong desire for an immediately available cell therapy option however, development of "off-the-shelf" T cells is challenging. Despite the success of autologous chimeric antigen receptor (CAR)-T cells, barriers to a more widespread use of this potentially curative therapy include manufacturing failures and the high cost of individualized production.

All of the tents for the whole crew were supplied by another sponsor for the trip, a 75-year-old canvas tent company called SpringBar Tents. I assigned myself a place to lay my head for the next 4 nights. The inside of my SpringBar home for five days. We used a lot of different gear that was supplied to us by many different brands, which I’ll touch on.īut, after going through what I’ve gathered and the photos I’ve taken, as well as going through all of the amazing photos that Eric and his assistant Emma took, I decided that first, we should let the photos do the talking. I saw several new, up-and-coming companies that were local to Colorado. The topography of the area varies drastically, from winding gravel roads through forests to summiting treeless, alpine passes then back down to the scrubby high desert and riding through huge valleys of hay fields.Īll of which provided endless views and dramatic landscapes that were, quite simply, jaw-dropping. There are approximately 1200+ miles of unpaved and groomed gravel roads throughout Gunnison County. The trip was to highlight the amazingly diverse area of Gunnison and show off some of the stellar gravel riding that can be done there. The trip was put together by polar explorer Eric Larson, his wife, and outdoor industry vet, Maria Hennessy of Smak Strategies, working alongside the Gunnison Crested Butte Tourism and Prosperity Partnership (TAPP). In mid-September, I was invited to camp and ride bikes in Gunnison County, Colorado for 5 days. It can also be a great way to set up base camp for epic gravel adventures. Looking for a gravel adventure? Maybe one where you can experience the great outdoors without having to carry all your gear on your bike? Well, glamping (glamorous camping) isn’t just for celebrities and casual campers. The short answer, for me at least, is that it does. Ask questions, and your virtual assistant will answer, whether it’s to tell you about your day or to turn down your smart light bulbs – but does adding a touchscreen really help? After all, surely the point of a voice assistant is to cut out the need for fingers and screens? You probably know the drill with smart speakers now. You’ll want to take this into account when you decide where to place it but unless you live in a greenhouse, this shouldn’t be a huge issue. That’s a pretty modest boost, and mostly gobbled up by the size increase, meaning that things look pretty similar to how they did on the older model: things aren’t hugely sharp but the colours look rich and largely accurate and that’s pretty much what you need for something that will be viewed from further away than your average phone or tablet. The extra size also calls for a resolution boost, going from 1,024 x 600 on the older model to 1,280 x 800 on the new version. The short digest of this section is that the Nest Hub Max looks like the Google Home Hub, only a bit bigger, with the original’s 7in screen gaining an extra 3in. Google Nest Hub Max review: Design and display The Lenovo Smart Display, for example, is available in both 8in and 10in configurations and the larger one currently sells for £180. There are other Google Assistant-imbued smart screens. The big rival for the Google Assistant is, of course, Amazon Alexa and, while the Echo Show 5 sells for £79, the 10in Echo Show sells for £219 – in other words, it’s a dead heat between the two. That kind of price increase is, however, pretty standard. How much more expensive? Well, while the original Google Home Hub went for £139, the Google Nest Hub Max will sell for £219. Google Nest Hub Max review: Price and competition Otherwise, it’s pretty much the same deal, only more expensive. The Next Hub Max gets a larger 10in 1,280 x 800 display, beefier sound by dint of having space for a more substantial speaker, and a camera for video chats and facial recognition. Google’s first smart screen – the Google Home Hub – was a 7in device with a 1,024 x 600 panel. You can also ask for YouTube videos, which is handy for a kitchen-bound screen if you like to see recipes and so on. You can ask Google Assistant the same things here as you would with a Google Home – the weather, how the traffic is, the latest news – only now it also provides visuals. The idea is that you take the digital assistant from the smart speaker and add a screen, letting it communicate in pictures as well as words. At this time one low center was moving eastward near Quantico, Virginia, while another was also moving eastward near Danville, Virginia. Snow had spread from southern New Jersey, beginning at 10:45 AM at Newark and 11:03 AM at New York City. Precipitation fell as rain over southern Maryland and southern Virginia, while snow was falling over the remainder. By 7:30 AM EST on the 18 th, the area of snowfall attending the developing southern low centers over eastern Kentucky and southern West Virginia had enlarged to cover the Ohio Valley and had spread eastward over the Appalachians to cover most of the Mid-Atlantic states. US Weather Bureau Daily Weather Map at 1:30 AM EST on 19 March 1956.Īll of this set the stage for the biggest storm, which occurred from March 18-20 th. Snowfall at Blue Hill Observatory measured 12.9 inches, and a peak wind gust to 78 miles per hour was recorded. In just 12 hours the intense storm moved from a position off the coast near Atlantic City to 380 miles east of Boston. It deposited 14 inches of new snow at Albany, New York., 6 inches at Hartford, Connecticut, and 10 inches at Concord, New Hampshire, New York City and Boston. This storm was a nearly perfect example of the rapid development of a coastal storm. By 7:30 AM EST, March 17, the storm was well out to sea some 380 miles east of Boston. Snow and strong winds covered the Northeastern states and gales continued on the New England coast. By 1:30 AM EST, March 17, the lowest pressure was 970 mb just east of Nantucket. Snow had now spread over all of the North Atlantic states, attended by strong winds with gales on the coast. By 7:30 PM EST on March 17, the low center in eastern Kentucky had entirely filled and the secondary low had formed and deepened to 984 mb just off Atlantic City, New Jersey The pressure at Atlantic City fell 25 mb in just 12 hours, indicating the explosive nature of the cyclogenesis that took place. An area of 3-hourly pressure falls of 4 to 5 mb, concentrated in eastern North Carolina and southeastern Virginia, strongly indicated a secondary development on the coast. A warm front lay along the Carolina coast and extended eastward north of Bermuda. By this time the typical pattern of development was evident. This storm brought widespread heavy rains in the southeastern states and snow through the Ohio Valley eastward to southern New Jersey. Meanwhile, a wave, which developed in the western Gulf of Mexico on the trailing polar front, had deepened and moved northeastward to eastern Kentucky. The North Atlantic states had been flooded with cold air due to eastward passage of a 1032-mb high from the Great Lakes region to Maine. The next storm was just two days later.īy 7:30 AM EST on Friday, March 16, 1956, the stage was set for the development of a coastal low that typically produces heavy snow and high winds in the Northeast. The storms started with a rather typical coastal development on March 14 th, which left 2.7″ of snowfall at Blue Hill Observatory. What was remarkable was that there were three snowstorms in March 1956 in a period of just 11 days from the 14 th to the 24 th. Of the three major snowfalls in March 1956 the storm from March 18-20 th was the most significant with upwards of 20 inches falling along the Northeast corridor and much of Southern New England. Including additional snow in April, the winter of 1955-56, with a total of 106.8 inches, was the third snowiest on record up to that time, and it is currently the eighth snowiest on the Observatory’s 133-year record. By the end of the month, the snow depth had peaked at 25 inches, and three 12-inch plus snowstorms had blanketed the Northeast bringing the monthly total at Blue Hill to a March record of 52.0 inches. March 1956 started with 5 inches of snow on the ground at Blue Hill Observatory. In New England, winter can still rule in March and heavy snows can occur even as late as April. The strong snowstorms in less than two weeks this March bring back memories of the big snows of March 1956. US Weather Bureau Daily Weather Map at 1:30 AM EST on 17 March 1956.
